- Experienced in generating conditional knockout mice.
- Experienced in studying the function of a putative tumor suppressor in T cell development from T-cell-specific conditional knockout mice.
- Experienced in studying the function of a putative tumor suppressor in hepatocyte development and liver development from liver-specific conditional knockout mice.
- Familiar with mouse in vivo transgenic or chemical carcinogenesis models of cancer research: skin cancer (DMBA and TPA two-stage carcinogenesis), liver cancer (DEN or aflatoxin B1 carcinogenesis), lung cancer (transgenic k-ras carcinogenesis), breast cancer (transgenic Wap-Tg, mmtv-Her or MNU carcinogenesis) and prostate cancer (transgenic TRAMP carcinogenesis).
- Experienced in using small interfering RNA (siRNA) to down regulate gene expression in cell lines and chicken embryo.
- Experienced with using DNA Microarray and Quantitative RT-PCR to evaluate gene expression in mammalian cancer research.
- Proficient in molecular cloning and cell culture techniques.
- Proficient at using the yeast two-hybrid system to screen new targets.

Major research experience
- Isolation and characterization of cancer stem-like cells in oral cancer: Enrichment of cancer stem-like cells from oral cancer cells or primary oral cancer tissues by serum-free medium plus bFGF and EGF growth factors or side population cells (Hoechst33258 dye exclusion). Establishment of radioresistant oral cancer cells by progressive selection.
- Generation of different tissue specific Tid1 knockout mice by breeding Tid1-floxed mice with individual Cre transgenic mice (Lck-Cre, CD4-Cre, Alb-Cre, K14-Cre, WAP-Cre, mmtv-Cre, Syn-Cre or MX-Cre) to understand the function of DNAJA3 (Tid1) on different tissues: total RNA preparation, DNA microarray, RT-PCR, MTS or CFSE cell proliferation, flow cytometery, MTT assay, coimmunoprecipitation, Western blot, Southern blot, immunochemical staining (confocal microscopy), histology assay, mouse anatomy, blastocyst outgrowth, T cell isolation, hepatocyte isolation and keratinocyte isolation.
- Conditional transgenic mice establishment to study the function of DNAJA3 (Tid1) as a tumor suppressor: Genomic database search, bioinformative search, design of gene targeting vector, construction of gene targeting plasmids, screening of gene of interest from BAC genomic library, primary mouse embryonic fibroblast (MEF) and embryonic stem cell (ES) culture, DNA electroporation on ES cells, stable undifferentiated ES cell line establishment, genomic DNA preparation of ES cell, screening of homologous recombination on mouse ES cell by Southern hybridization and genomic PCR, targeted mice germ lines selection, and adenovirus or retrovirus gene delivery.
- Down regulation of genes expression upon inhibition of BMK1 kinase by RNA interference in cancer cell lines to elucidate the function of BMK1 on tumorigenesis: Utility of siRNA technique to down regulate the posttranscriptional mRNA of BMK1 kinase, MTT assay, total RNA preparation, DNA microarray, flow cytometery, coimmunoprecipitation and Western blot.
- Yeast two-hybrid to screen the erbB2 interacting proteins involving in the signal transductions of breast cancer cells: Cloning of genes of interest for yeast two-hybrid screening, yeast two-hybrid screening on high throughput 96- well format, DNA transfection of mammalian cell line, stable cell line establishment, rabbit polyclonal antibody generation, total mRNA extraction, MTT assay, b-galactosidase activity assay, adenovirus expression construct preparation and establishment, Western blot, RT-PCR, construction of cDNA library, deletion or site-directed mutagenesis, peptide mapping, phosphopeptide mapping, coimmunoprecipitation and cis or trans reporting assay (luciferase activity assay).
- Cloning, expression, purification and characterization of a silver binding protein (SilE) from the genes of Salmonella: Cloning of SilE over expression strain by PCR, purification of SilE by ammonium sulfate precipitation, cation exchange chromatography and gel filtration or HPLC, peptide sequencing, circular dichroism (CD) to study the secondary structure change after the binding of Ag+ to SilE, inductive coupling plasma (ICP), atomic absorption spectroscopy and mass spectroscopy to study the stoichiometry of Ag+ binding to SilE, NMR (proton or HSQC) to study the change of imidazole groups of histidine residues of SilE that are responsible for Ag+ binding and replacement of tryptophan on phenylalanine of SilE by site-directed mutagenesis to study the binding constant of SilE to Ag+ through fluorescence spectroscopy.
- Cloning, expression and purification of a DNA binding protein (CadC, renamed CadR) on the cadmium resistance mechanism in bacteria (Bacillus, Staphyl and methicilline resistant Staphyl): Cloning of GST-cadC fusion strain by PCR, gel retardation, probe labeling, mass growth of bacteria by fermentation, GST-CadC purification by affinity chromatography, anion exchange chromatography and gel filtration.
- Crystallization of mutant protein of CheY, a regulatory protein of the two-component system in E. coli: Cloning of expression plasmid, purification of over expressed protein by gel filtration (FPLC), affinity chromatography, ion exchange chromatography, SDS-PAGE, crystallization by hanging-drop and data collection on X-ray diffraction.
- Selection of transformed cell line by Bovine Papilloma Virus: Cell line transfection, cell line establishment from a single colony on soft agar.
- Purification and characterization of Glutathione S-transferase p from an arsenic-resistant Chinese hamster ovary cell line: Cell culture, GST purification through affinity chromatography, 1D and 2D electrophoresis, characterization on enzymatic kinetics of GST-p by photospectroscopy, polyclonal antibody preparation against GST-p, Western blot, cytotoxicity assay by colony formation.
